Coiled Therapeutics PLC - London-based clinical-stage oncology company - Says it has successfully enrolled patients across two escalation cohorts for the clinical development of its lipid-based formulation of brain-penetrant small-molecule TACC3 inhibitor AO-252. One cohort will be given a twice-daily 120 miligram dose, the other 160mg once a day. "All patients are progressing as planned through the assessment period, in accordance with the study protocol," Coiled says. Explains that the new formulation "is designed to achieve more consistent, dose-proportional absorption," in light of the 80% clinical benefit rate signal it observed in twice-daily tablet dosing. The dosages "were selected to optimise systemic drug exposure based on encouraging clinical activity observed with the original tablet formulation." Coiled says enrolment marks further progress towards its planned phase 1/2 dose-expansion cohorts in ovarian and prostate cancer. It aims to enrol up to 30 patients with the new formulation by the end of 2026, and also continues to expect initial safety and pharmacokinetic data readouts by the end of 2026. Read More
